PfSPZ-LARC2 is a next-generation, genetically engineered malaria vaccine developed to provide high-level protection against *Plasmodium falciparum* infection with a single dose. The vaccine uses live, attenuated sporozoites of *P. falciparum* that have been genetically modified by deleting the Mei2 and LINUP genes, which are essential for the parasite's transition from the liver stage to symptomatic blood stage infection. This genetic attenuation causes late liver-stage arrest, meaning the parasites can infect liver cells but cannot progress to cause disease in humans. As a result, PfSPZ-LARC2 induces strong humoral and cellular immune responses without risk of breakthrough blood-stage infection[1][3][4][5]. Developed by Sanaria in collaboration with Seattle Children’s Research Institute’s Center for Global Infectious Disease Research (CGIDR), this third-generation vaccine aims to achieve at least 90% protection against malaria and is being evaluated in clinical trials across multiple countries[1][2].
