PfSPZ-LARC2 is an experimental malaria vaccine regimen consisting of live, infectious *Plasmodium falciparum* sporozoites (PfSPZ) administered via direct venous inoculation in conjunction with the long-acting antimalarial drug pyrimethamine. This approach, known as Chemoprophylaxis Vaccination (CVac) or Long-Acting Chemoprophylaxis (LARC), utilizes the drug to arrest the parasite's development at the late liver stage or early blood stage, thereby preventing clinical malaria while allowing the host's immune system to be exposed to a broad array of liver-stage antigens. This exposure induces potent, protective T-cell responses. Developed by Sanaria in collaboration with the University Hospital Tübingen, PfSPZ-LARC2 is designed to provide durable, high-level protection against malaria with a simplified dosing schedule compared to earlier CVac iterations.
