Overview

PG-102 is a novel bispecific Fc-fused protein agonist targeting both the glucagon-like peptide 1 receptor (GLP-1R) and glucagon-like peptide 2 receptor (GLP-2R). Developed by ProGen, it is designed for the treatment of obesity and type 2 diabetes. The drug acts as a dual GLP-1/GLP-2 agonist, aiming to improve body composition and glycemic control. Preclinical studies have shown that PG-102 offers superior efficacy in weight loss and metabolic improvement compared to other agents such as tirzepatide. Clinical trials indicate that subcutaneous administration of PG-102 results in significant weight reduction with a favorable safety profile—primarily mild gastrointestinal side effects typical for this class. The drug’s prolonged pharmacokinetic profile allows for flexible dosing intervals ranging from weekly to monthly injections. An oral version delivered via Rani Therapeutics’ RaniPill capsule (RT-114) has demonstrated bioequivalence to injectable forms in preclinical models[1][2][3][5][8].
