PG13-CEA_TCR is an experimental T-cell receptor (TCR) engineered T-cell therapy designed for the treatment of cancers expressing Carcinoembryonic Antigen (CEA). In this adoptive cell therapy approach, autologous T lymphocytes are genetically modified to express a murine T-cell receptor that specifically recognizes human CEA when presented on tumor cells in the context of the major histocompatibility complex (MHC). The mechanism of action involves these engineered T cells targeting and killing CEA-positive tumor cells. Early clinical investigations, such as a study conducted by the National Cancer Institute (NCI), have explored its use in patients with metastatic colorectal cancer. While demonstrating profound decreases in serum CEA levels and objective tumor regression in some patients, severe transient inflammatory colitis was observed as a dose-limiting toxicity.
