PG13-MAGE-A3 TCR9W11 is an autologous cell therapy in which a patient’s own peripheral blood lymphocytes are genetically engineered to express a high-avidity, modified T-cell receptor (TCR) that targets the melanoma-associated antigens MAGE-A3 and MAGE-A12. The therapy is designed for adoptive cell transfer immunotherapy in cancer patients whose tumors express these antigens. The engineered cells are infused back into the patient following lymphodepleting chemotherapy and supported with interleukin-2 to enhance their survival and activity. This approach aims to mediate tumor regression by redirecting the immune system against cancer cells expressing MAGE family antigens. Clinical development was led by the National Cancer Institute, primarily targeting metastatic melanoma and other neoplasms[1][2].
