PGA-AVE1642 is a **poly-l-glutamic acid polymer conjugate** of the humanized anti-insulin-like growth factor 1 receptor monoclonal antibody AVE1642, developed in preclinical research as an antitumor biologic for prostate cancer. The conjugate retains IGF-1R binding but appears to alter the antibody bio-nano interface and mechanism, increasing receptor-binding affinity, reducing IGF-1R internalization relative to native AVE1642, and inhibiting both phosphoinositide 3-kinase and mitogen-activated protein kinase signaling in prostate cancer models. In published preclinical work, PGA-AVE1642 showed enhanced antitumor activity versus unmodified AVE1642 in an orthotopic prostate cancer mouse model, with greater suppression of tumor-cell proliferation and angiogenesis.
