Overview

PGDM1400 is a broadly neutralizing monoclonal antibody (bNAb) targeting the apex of the HIV envelope glycoprotein trimer, specifically binding to the V2 region of gp120. It exhibits exceptional breadth and potency, with cross-clade neutralization coverage of approximately 83% at a median IC50 of 0.003 μg/mL. PGDM1400 is being investigated as an intravenous therapy for both treatment and prevention of HIV infection, often in combination with other bNAbs such as PGT121 and VRC07-523LS. The drug has demonstrated safety and tolerability in early-phase clinical trials, with most adverse events being mild or unrelated to the antibody itself. A long-acting variant, PGDM1400LS, has also been developed to extend plasma half-life for less frequent dosing[1][5][7].
