Overview

**PGG-PTX** is a water-soluble nanoparticle formulation of paclitaxel, created by covalently linking paclitaxel (PTX) to poly(L-γ-glutamylglutamine) acid (PGG) via ester bonds[1][4]. This modification enhances water solubility, promotes self-assembly into nanoparticles of 12–20 nm, and allows for superior tumor uptake through prolonged circulation and tumor retention compared to free PTX or other polymer–paclitaxel formulations. The PGG-PTX conjugate releases paclitaxel slowly in plasma but accumulates and delivers more active drug within tumor tissue, prolonging its exposure and efficacy while reducing systemic toxicity. Preclinical studies have shown improved antitumor activity and a higher maximum tolerated dose versus established paclitaxel formulations[1][3][4]. Developed as an antineoplastic agent, its primary indications are various solid tumors, including non–small cell lung cancer[2][4].
