Overview

PGN 9856-isopropyl ester is a highly potent, selective, and long-acting small molecule agonist of the prostaglandin E2 (EP2) receptor. It is under development primarily as a next-generation ophthalmic agent for the treatment of glaucoma, notable for its ultra-long-acting ocular hypotensive effects, with single dosing producing intraocular pressure reductions lasting at least 5 days in preclinical primate models. The compound demonstrates high selectivity and high affinity for the EP2 receptor, minimal activity for other prostanoid receptors, and exhibits anti-inflammatory and tocolytic properties. Topically, it can be administered via either conventional eye drops or periorbital dermal application with a roller-ball device, offering potential for improved patient compliance through noninvasive, extended-duration delivery[1][2][3][4][5][7].
