Overview

PGN635 is a fully human monoclonal antibody that targets phosphatidylserine (PS), a phospholipid that is normally sequestered on the inner leaflet of the cell membrane but becomes exposed on the outer surface of tumor vascular endothelial cells and tumor cells in response to oxidative stress. Developed through a collaboration between Affitech A/S, UT Southwestern Medical Center, and Peregrine Pharmaceuticals, PGN635 acts as an immunotherapy and vascular-targeting agent. By binding to PS, it reverses PS-mediated immunosuppression and triggers immune-mediated destruction of the tumor vasculature and cancer cells. It has been investigated in preclinical and early clinical models for both therapeutic applications (e.g., prostate cancer, breast cancer, glioma) and as a molecular imaging agent when conjugated to contrast agents or nanoparticles for MRI.
