Overview

PGT121 is a recombinant human IgG1 monoclonal antibody and a second-generation broadly neutralizing antibody (bNAb) that targets a V3 glycan-dependent epitope on the HIV-1 envelope glycoprotein (Env). It potently neutralizes diverse HIV-1 strains by binding to complex-type N-glycans in the V3 region of gp120, thereby inhibiting viral entry into host cells. Structural studies show that it blocks gp120 binding to the CD4 receptor, preventing infection of new cells. In preclinical and clinical studies, PGT121 has demonstrated strong antiviral activity, including reducing viral load in animal models and delaying viral rebound after antiretroviral therapy interruption. The drug is under development for both treatment and prevention of HIV infection, with ongoing Phase 2 clinical trials evaluating its safety and efficacy as monotherapy or in combination regimens[1][2][3][5].
