Overview

PH-894 is a self-delivering small interfering RNA (siRNA) compound developed using Phio Pharmaceuticals' INTASYL technology. It specifically targets and silences the BRD4 gene, a key regulator of gene expression involved in cancer progression and immune evasion. PH-894 exerts a dual mechanism of action by both enhancing the activation and proliferation of immune cells (such as T cells and natural killer [NK] cells) and making tumor cells more susceptible to immune-mediated killing. In preclinical studies, PH-894 has demonstrated the ability to increase tumor cell immunogenicity—such as upregulating MART-1 in melanoma—leading to improved recognition by T cells, while also directly inducing apoptosis in various human cancer cell lines. The compound is being developed primarily for use in adoptive cell therapies (including NK cell-based immunotherapies) and for local treatment of solid tumors such as melanoma, with the aim of providing effective antitumor activity while minimizing systemic toxicity commonly seen with small molecule BRD4 inhibitors[1][2][3][5][6][7].
