WuXi Biologics' pH-dependent FolR1 T cell engager is a bispecific antibody candidate designed to target Folate Receptor Alpha (FolR1) and CD3. It utilizes a pH-sensitive binding mechanism to selectively target the acidic tumor microenvironment (TME) while remaining inactive in normal tissues where pH is neutral. This design aims to mitigate on-target, off-tumor toxicity, a significant challenge for FolR1-targeted therapies due to the antigen's expression on the luminal surfaces of healthy tissues such as the kidney, lung, and intestine. In preclinical studies, the candidate demonstrated potent cytotoxicity against FolR1-positive tumor cells at acidic pH and showed significant tumor growth inhibition in ovarian cancer and non-small cell lung cancer (NSCLC) models. Exploratory toxicology studies in cynomolgus monkeys indicated a superior safety profile compared to conventional FolR1 T-cell engagers.
