Overview

Ph-HH phage is a genetically engineered filamentous fd phage developed as a tumor-targeting immunotherapeutic "bionanofibre." It is modified to display two distinct peptides: one that binds to the extracellular domain of Programmed cell death-ligand 1 (PD-L1) and another that specifically targets melanoma tissues and cancer cells. Upon intravenous administration, the phage selectively homes to the tumor microenvironment and binds to PD-L1 on cancer cells, thereby blocking the PD-1/PD-L1 checkpoint pathway. This targeted approach is designed to stimulate anti-tumor immunity while minimizing the systemic toxicities, such as body weight loss and organ abnormalities, often associated with conventional non-targeted PD-L1 monoclonal antibodies. The phages are cost-effectively produced through bacterial replication and can be adapted to target various immune checkpoints and cancer types.
