Ph-WG is a genetically engineered filamentous fd phage developed as a tumor-targeting immunotherapeutic "bionanofibre." It is designed to overcome the lack of tumor-homing capabilities in conventional PD-L1 blockers. The phage is modified to display two specific peptides: one that binds to the extracellular domain of Programmed cell death-ligand 1 (PD-L1) and another that targets melanoma tissues and cancer cells. Upon intravenous administration, the Ph-WG phage homes to the tumor site and binds to PD-L1 on the surface of cancer cells, effectively blocking the PD-1/PD-L1 inhibitory checkpoint. This blockade reactivates the anti-tumor immune response while minimizing systemic side effects. The phage-based platform is cost-effective to produce via bacterial replication and can be adapted to target various cancer types and immune checkpoints.
