Overview

PH033 is an investigational targeted protein degrader (TPD) being developed by Suzhou Puzheng Pharma (also known as Puhe Biopharma) for the treatment of solid tumors. It is designed as a pan-RAS degrader, utilizing either a Proteolysis-Targeting Chimera (PROTAC) or molecular glue approach to induce the ubiquitination and subsequent proteasomal degradation of various RAS isoforms, including KRAS, NRAS, and HRAS. By targeting the degradation of these key oncogenic drivers rather than merely inhibiting their signaling, PH033 aims to provide a more robust and comprehensive therapeutic response in cancers driven by RAS mutations, which are among the most common genetic alterations in human malignancies, particularly in pancreatic, colorectal, and lung cancers.
