**PH46A** is a first-in-class **1,2-indane dimer** with potent anti-inflammatory and immune-modulating activity, specifically the *N*-glucamine salt of the *(1S,2S)-enantiomer* of PH46. Developed as a novel therapeutic for **inflammatory and autoimmune diseases**, particularly **inflammatory bowel disease (IBD)**, it demonstrates efficacy in murine models of colitis (DSS and IL-10^−/−^*), reducing colon cytokines (TNF-α, IL-6, IL-1β) and histological damage. PH46A inhibits **5-lipoxygenase (5-LOX)**, nitric oxide (NO) production, and cytokines (IL-6, TNF-α, IL-8, IFN-γ) in vitro without cytotoxicity, dissociating in solution to its active PH46 form.[1][2]
