Phacetoperane is a psychostimulant small molecule originally developed by Rhône-Poulenc in the 1950s. It is structurally a reverse ester of methylphenidate. The (R,R) enantiomer, known as levophacetoperane (also referred to as NLS-3), has been investigated for its therapeutic potential. Historically, phacetoperane was marketed as Lidepran® in Canada and Europe from 1959 to 1967 for the treatment of obesity and depression. It has also been used as an anorectic and a wakefulness-promoting agent for narcolepsy. More recently, NLS Pharmaceutics has been developing NLS-3 (levophacetoperane) as a potential treatment for Attention-Deficit/Hyperactivity Disorder (ADHD) and narcolepsy, aiming to offer a safer and more potent alternative to existing stimulants.
