Overview

PHAD is a fully synthetic structural analog of monophosphoryl lipid A (MPLA), designed as a vaccine adjuvant. It mimics the immune-stimulating effects of bacterial-derived MPLA but with higher purity and typically lower toxicity. As a Toll-like receptor 4 (TLR4) agonist, PHAD activates innate immunity by stimulating TLR4 on immune cells. This activation leads to the production of proinflammatory cytokines and enhances both CD4+ effector and memory CD8+ T cell responses. The use of PHAD as an adjuvant can enhance immunogenicity and skew the immune response toward a Th1-type profile, which is particularly effective against viruses and intracellular bacteria. It has been administered to over 1000 human subjects in clinical research without serious adverse events and is available for vaccine development and commercial manufacturing[1][2][3][4].
