Drug intelligence / Profile preview
PHB1-NES peptide is a synthetic peptide designed to inhibit the nuclear export of prohibitin 1 (PHB1). PHB1 is a multifunctional protein that shuttles between the nucleus, cytoplasm, and mitochondria, and its subcellular localization dictates its function. Under stress conditions, such as ethanol exposure or camptothecin treatment, PHB1 interacts with exportin 1 (CRM1), leading to its nuclear depletion. By mimicking the nuclear export signal (NES) of PHB1, the PHB1-NES peptide competitively blocks the interaction between PHB1 and CRM1, thereby preventing the nuclear export and depletion of PHB1. In preclinical models, preservation of nuclear PHB1 by PHB1-NES peptide protects against alcohol-associated liver disease (ALD) and ethanol-mediated sensitization to colorectal liver metastasis (CRLM).
