Overview

PHD2-targeting oligonucleotide is a chemically modified short synthetic hairpin RNA (sshRNA) developed by SomaGenics, Inc. in collaboration with Stanford University for the treatment of diabetic wound healing. The therapeutic works by silencing the expression of prolyl hydroxylase domain-containing protein 2 (PHD2), an enzyme responsible for the degradation of hypoxia-inducible factor-1α (HIF-1α). By inhibiting PHD2, the compound stabilizes HIF-1α levels even under normoxic conditions, which in turn activates a pro-angiogenic and pro-regenerative gene program. This leads to enhanced cellular motility, proliferation, and new vessel formation, addressing the impaired healing characteristic of chronic diabetic wounds. The oligonucleotide is formulated for local delivery using layer-by-layer (LbL) nanotechnology applied to a polymer-coated mesh, allowing for sustained release at the wound site.
