Drug intelligence / Profile preview
PHDP5 (Pleckstrin Homology Domain Peptide 5) is a synthetic dodecapeptide derived from the pleckstrin homology domain of dynamin 1. Developed by researchers at the Okinawa Institute of Science and Technology Graduate University (OIST), PHDP5 acts as a microtubule-dynamin binding inhibitor. In Alzheimer's disease and other tauopathies, the accumulation of soluble tau leads to microtubule over-assembly, which abnormally binds and depletes cytosolic dynamin, thereby impairing synaptic vesicle endocytosis and synaptic transmission. By inhibiting the interaction between microtubules and dynamin, PHDP5 restores the availability of cytosolic dynamin, rescuing endocytic and synaptic dysfunction. Administered intranasally, the peptide has demonstrated the ability to cross the blood-brain barrier, reach the hippocampus, and reverse spatial learning and memory deficits in preclinical mouse models of Alzheimer's disease.
