Overview

PhenDC3 is a highly potent, selective bisquinolinium small-molecule stabilizer of G-quadruplex (G4) DNA and RNA. Developed by the Teulade-Fichou group at the CNRS and Institut Curie, PhenDC3 binds to telomeric and promoter G-rich sequences with nanomolar affinity, altering G4 topology through intercalation. In research settings, it is widely used as a benchmark G4-stabilizing probe to study G4-dependent biological processes, including transcription inhibition, telomere dysfunction, replication stalling, and the activation of the cGAS-STING innate immune pathway via micronuclei formation. It has also demonstrated potential in preclinical models of cancer, fungal infections, and sepsis-associated vascular injury.
