Phenindione

Phenindione is a synthetic oral anticoagulant that functions as a Vitamin K antagonist. It interferes with the formation of vitamin K-dependent clotting factors (II, VII, IX, and X) in the liver by inhibiting vitamin K reductase, leading to depletion of reduced vitamin K (vitamin KH2). This action reduces the blood's ability to clot. It is prescribed for thromboembolic disorders such as deep vein thrombosis, pulmonary embolism, and to reduce the risk of stroke in patients with atrial fibrillation. While it has actions similar to warfarin, it is less commonly used today due to a higher incidence of severe adverse effects, including hypersensitivity reactions.
