Overview

Phenoxybenzamine is a non-selective, irreversible alpha-adrenergic antagonist used primarily to manage and treat hypertension and excessive sweating associated with pheochromocytoma, a tumor of the adrenal gland. It works by covalently binding to both alpha-1 and alpha-2 adrenergic receptors, leading to long-lasting blockade of these receptors. This action causes vasodilation by relaxing vascular smooth muscle, resulting in lowered blood pressure. Phenoxybenzamine is administered orally and has a prolonged duration of effect due to its irreversible receptor binding[1][2][3][5][6]. The drug does not affect the parasympathetic system.
