Overview

Phentermine + pramlintide is an investigational combination therapy for obesity and weight management. Phentermine is a sympathomimetic amine anorectic that acts as an appetite suppressant, primarily through the release of norepinephrine in the hypothalamus. Pramlintide is a synthetic analogue of amylin, a hormone co-secreted with insulin by pancreatic beta cells, which regulates satiety and slows gastric emptying. The combination targets distinct regulatory pathways involved in energy balance—phentermine reduces appetite via central nervous system stimulation, while pramlintide enhances satiety and delays gastric emptying peripherally. Clinical studies have shown that this combination produces greater weight loss than either agent alone or placebo[1][3]. The primary indication under investigation has been obesity/weight loss.
