Overview

Phenylpropanolamine is a sympathomimetic small molecule that was widely used as a decongestant and appetite suppressant. It acts primarily as an indirect agonist of adrenergic receptors by promoting the release of norepinephrine, leading to vasoconstriction and reduced nasal congestion. It also inhibits norepinephrine reuptake at synaptic junctions. Phenylpropanolamine was commonly found in over-the-counter cold, allergy, and weight loss products but has been withdrawn from the market in many countries due to its association with an increased risk of hemorrhagic stroke, particularly in women. The drug is structurally related to amphetamine, ephedrine, and pseudoephedrine[1][3][5][6].
