PHER-02 is a next-generation antibody-drug conjugate (ADC) targeting Human Epidermal Growth Factor Receptor 2 (HER2), developed by Pheron Therapeutics. It utilizes a proprietary site-specific conjugation technology (PH-Linker) designed to ensure a stable and uniform drug-to-antibody ratio (DAR), which aims to enhance therapeutic efficacy while minimizing systemic toxicity. PHER-02 is engineered to deliver a potent cytotoxic payload, typically a topoisomerase I inhibitor, directly to HER2-expressing tumor cells. The drug is primarily being developed for the treatment of HER2-positive solid tumors, including breast and gastric cancers, and is specifically designed to address unmet needs in patients with low HER2 expression (HER2-low) or those who have developed resistance to existing HER2-targeted therapies such as trastuzumab emtansine.
