Drug intelligence / Profile preview
Phesgo is a fixed-dose combination biologic therapy containing two HER2-targeting monoclonal antibodies, pertuzumab and trastuzumab, along with hyaluronidase-zzxf, an enzyme that facilitates subcutaneous administration. Developed by Roche and its subsidiary Genentech, Phesgo is FDA-approved for the treatment of HER2-positive breast cancer. In the context of prostate cancer, the Mayo Clinic is investigating Phesgo in the Phase II TraPPer trial (NCT05730712) in combination with enzalutamide for patients with enzalutamide-refractory metastatic castration-resistant prostate cancer (mCRPC). The study aims to determine if dual HER2 blockade can overcome resistance to androgen receptor signaling inhibitors in this patient population. The subcutaneous delivery of Phesgo offers a more convenient administration route compared to traditional intravenous infusions of its individual components.
