Overview

PHIP shRNA is a research-stage RNA interference (RNAi) therapeutic candidate designed to silence the expression of the Pleckstrin Homology domain-Interacting Protein (PHIP). PHIP is a multi-domain protein that acts as a molecular scaffold, promoting cancer progression and metastasis through the activation of the IGF1R/AKT signaling pathway. Overexpression of PHIP is associated with poor prognosis in several malignancies, including melanoma, triple-negative breast cancer, and non-small cell lung cancer. By utilizing short hairpin RNA to knock down PHIP expression, this modality inhibits tumor cell proliferation, colony formation, and invasive potential. Developed primarily in academic settings such as the California Pacific Medical Center Research Institute and UCSF, PHIP shRNA serves as both a tool for target validation and a potential lead for future gene therapy applications.
