Phloroglucinol is a non-atropinic, musculotropic antispasmodic agent used primarily for the symptomatic treatment of pain related to functional disorders of the gastrointestinal tract, biliary tract, and urinary tract, as well as for acute spasmodic pain in gynecology. Unlike anticholinergic antispasmodics, phloroglucinol acts directly on smooth muscle fibers, inhibiting spasms without inducing the typical side effects associated with atropine-like drugs, such as dry mouth, tachycardia, or blurred vision. Its mechanism of action involves the inhibition of voltage-gated calcium channels and potentially the modulation of other intracellular signaling pathways in smooth muscle cells. It is frequently utilized in the management of renal colic, biliary colic, and dysmenorrhea, and is also used in obstetrics to facilitate cervical dilation during labor.
