Overview

pHM-GM-CSF is an investigational **bicistronic plasmid DNA cancer vaccine** encoding a truncated human epidermal growth factor receptor 2 antigen and granulocyte-macrophage colony-stimulating factor. Intramuscular administration is intended to induce HER2-directed cellular and humoral antitumor immunity while locally expressed GM-CSF enhances antigen presentation and immune priming. It was evaluated as the DNA-prime component of a heterologous pHM-GM-CSF/Ad-HM prime-boost regimen in patients with HER2-expressing stage III-IV metastatic breast cancer; the phase 1 study reported mainly grade 1 toxicities and no serious drug-related adverse events.
