Overview

PHN131 is an oral soft-capsule formulation of the mixed opioid agonist–antagonist nalbuphine hydrochloride being developed for the treatment of moderate-to-severe acute postoperative pain, particularly after hemorrhoidectomy and other surgeries. As a κ-opioid receptor agonist and μ-opioid receptor antagonist, nalbuphine provides analgesia with a lower risk of respiratory depression, hemodynamic instability, and addiction liability compared with traditional μ-opioid agonists such as morphine. In randomized, double-blind, placebo-controlled studies in post-hemorrhoidectomy patients, PHN131 60 mg soft capsules significantly reduced pain scores, delayed time to rescue analgesia, and decreased cumulative diclofenac use, with mainly mild-to-moderate adverse events, supporting its potential as an effective and well-tolerated oral analgesic option for postoperative pain management.[1][5][7][9]
