Overview

Photrexa is a sterile, buffered ophthalmic solution containing riboflavin 5’-phosphate (vitamin B2) used as a photoenhancer in combination with the KXL System for corneal collagen cross-linking. Its primary indications are the treatment of progressive keratoconus and corneal ectasia following refractive surgery. The mechanism involves topical application of Photrexa to the de-epithelialized cornea followed by ultraviolet A (UVA) irradiation. Riboflavin acts as a photosensitizer, generating singlet oxygen upon UVA exposure, which induces cross-linking of collagen fibers in the cornea. This process strengthens and stabilizes the corneal structure to slow or halt disease progression[1][2][3][4][6]. Photrexa was first approved in 2016[2].
