The PHOX2B epitope-encoded mRNA-LNP vaccine is an investigational cancer vaccine designed as a 'CAR boosting' strategy to enhance the persistence and anti-tumor potency of peptide-centric chimeric antigen receptor (PC-CAR) T cells. The vaccine consists of nucleoside-modified mRNA encoding a 9mer peptide monomer derived from the PHOX2B oncoprotein, encapsulated in the FDA-approved SM-102 lipid nanoparticle (LNP). PHOX2B is a major neuroblastoma oncoprotein, and this vaccine specifically targets the epitope presented in the context of HLA-A*24:02. By treating monocyte-derived dendritic cells (moDCs), the vaccine induces high-level antigen presentation and upregulates T cell costimulatory ligands such as CD80/86 and CD40. This process stimulates the proliferation, polyfunctionality, and memory formation of PC-CAR T cells, helping to overcome the hostile tumor immune microenvironment and poor persistence typically associated with adoptive cellular therapies in solid tumors like neuroblastoma.
