Overview

PHP-303 is a novel, orally administered small molecule inhibitor of neutrophil elastase (also known as leukocyte elastase or ELA2). Neutrophil elastase is a serine protease released by activated neutrophils and plays a key role in tissue destruction and inflammation, particularly in the lungs. By selectively inhibiting neutrophil elastase, PHP-303 aims to reduce inflammatory damage associated with diseases such as alpha 1-antitrypsin deficiency (AATD) and nonalcoholic steatohepatitis (NASH). The drug has been developed by pH Pharma, Akari Therapeutics, and Bayer HealthCare. Its primary clinical focus has been on AATD, where it has reached Phase 2 trials; it was previously investigated for bronchiectasis but development for that indication was discontinued[1][2][3][4][5].
