Overview

PHST001 is a humanized anti-CD24 monoclonal antibody developed by Pheast Therapeutics as a novel macrophage checkpoint inhibitor for the treatment of advanced solid tumors. It is designed to block CD24, a cell surface protein highly expressed on many human cancers—including ovarian and triple-negative breast cancer—that interacts with the macrophage receptor Siglec-10 to suppress phagocytosis and enable tumor immune evasion. By binding all glyco-variants of CD24 with high affinity and specificity, PHST001 prevents its interaction with Siglec-10, thereby promoting macrophage-induced phagocytosis of cancer cells and stimulating an innate immune response against tumors. Preclinical studies have shown that PHST001 induces robust antitumor activity across multiple cancer types without inducing cytokine release or immune-mediated toxicity. The drug is currently being evaluated in a Phase 1 clinical trial for patients with advanced relapsed or refractory solid tumors[1][2][3][4][5][6][7][8].
