PhtD vaccine is a protein-based pneumococcal vaccine candidate targeting Streptococcus pneumoniae. The active component, polyhistidine triad protein D (PhtD), is a conserved surface adhesin found in all pneumococcal strains and plays a role in bacterial adhesion and immune evasion by binding zinc and inhibiting complement deposition[2][1]. Vaccination with PhtD elicits functional antibodies that can block bacterial adhesion and provide protection against infection[1][2]. The C-terminal region of PhtD is highly immunogenic. Clinical studies have evaluated both free and conjugated forms of the antigen, sometimes combined with other proteins such as detoxified pneumolysin (dPly), showing acceptable safety profiles and robust immune responses in adults[5]. The primary indication under investigation is prevention of invasive disease caused by Streptococcus pneumoniae.
