**PI3Kδ-downregulated CD38-CAR-T** is a preclinical, nanobody-based anti-CD38 CAR-T-cell construct engineered with a lentiviral vector encoding short hairpin RNA against phosphatidylinositol 3-kinase delta. The CAR directs T-cell cytotoxicity toward CD38-expressing acute myeloid leukemia cells, while PI3Kδ knockdown is intended to improve CAR-T-cell functional properties and reduce inflammatory cytokine release. In reported AML cell-line, primary AML-cell, and mouse studies, the construct retained antileukemic activity and improved survival versus untreated controls; PI3Kδ-downregulated cells showed lower IL-2, interferon-gamma, and tumor necrosis factor release and better mouse survival than the corresponding CD38-CAR-T construct.
