Piasclédine

Piasclédine is a symptomatic slow-acting drug for osteoarthritis (SYSADOA) containing a specific 1:2 ratio of avocado and soybean oil unsaponifiables (ASU). Developed and manufactured by Expanscience, it is primarily indicated for the symptomatic treatment of osteoarthritis, particularly affecting the knee and hip joints. The drug's mechanism of action involves both chondroprotective and anti-inflammatory pathways; it stimulates the synthesis of extracellular matrix components like proteoglycans and type II collagen while simultaneously inhibiting the production of pro-inflammatory cytokines such as interleukin-1 (IL-1), IL-6, IL-8, and tumor necrosis factor (TNF). Additionally, it reduces the synthesis of prostaglandin E2 (PGE2), modulates the NF-kappa-B pathway, and decreases the expression of COX-2. Clinical efficacy is characterized by a delayed onset, with symptomatic improvement typically manifesting after 1 to 2 months of daily oral treatment.
