Overview

Piclidenoson (CF101) is an orally bioavailable, first-in-class small molecule agonist of the A3 adenosine receptor (A3AR). Developed by Can-Fite Biopharma, it is primarily being investigated for the treatment of moderate-to-severe plaque psoriasis and other inflammatory conditions. The drug's mechanism involves binding to A3AR, which is overexpressed in inflammatory cells, leading to the inhibition of pro-inflammatory cytokines such as interleukin-17 (IL-17) and interleukin-23 (IL-23), and inducing apoptosis in pathogenic keratinocytes. It has demonstrated a favorable safety profile in Phase 3 clinical trials (COMFORT study) compared to standard-of-care oral treatments like apremilast.
