PICRN1024 is a first-in-class, allosteric small molecule modulator of the eukaryotic translation initiation factor 4E (eIF4E), developed by PIC Therapeutics. It functions by targeting the Pre-Initiation Complex (PIC) to selectively modulate protein translation, specifically impacting the synthesis of key oncogenic drivers. The drug is primarily being developed for the treatment of estrogen receptor-positive (ER+) metastatic breast cancer, including cases that have acquired resistance to CDK4/6 inhibitors. Preclinical studies have demonstrated that PICRN1024 reduces the expression of ERα, Cyclin D1, and CDK4, and decreases phospho-Rb levels, leading to reduced cancer cell viability and the potential to overcome multiple drug resistance mechanisms.
