PICRN941 is an orally bioavailable, small molecule allosteric modulator of eukaryotic translation initiation factor 4E (eIF4E) developed by PIC Therapeutics. It is designed to target the Pre-Initiation Complex (PIC) to selectively modulate CAP-dependent protein translation, thereby reducing the expression of key oncogenes such as MYC, CDK2, CDK4, CCND1, and eIF4E itself. Preclinically, PICRN941 has demonstrated the ability to induce cell cycle arrest and apoptosis in estrogen receptor-positive (ER+) breast cancer cell lines and patient-derived organoids, offering a potential therapeutic approach to overcome resistance to standard-of-care therapies like CDK4/6 inhibitors, hormonal therapies, and PI3K inhibitors.
