Overview

**pictilisib + carboplatin + paclitaxel** is an experimental combination regimen consisting of pictilisib, an oral pan-class I phosphatidylinositol 3-kinase (PI3K) inhibitor, and two established chemotherapy agents, carboplatin and paclitaxel. Pictilisib inhibits the PI3K pathway, a key signaling axis frequently dysregulated in cancer, leading to reduced cellular proliferation, survival, and tumor growth. Carboplatin is a platinum-based alkylating agent that causes DNA crosslinking and apoptosis, while paclitaxel is a taxane that stabilizes microtubules, thereby disrupting mitotic division. This three-drug combination has been studied primarily in advanced non-small cell lung cancer (NSCLC) and other solid tumors to assess safety and preliminary antitumor activity, generally as a first-line or advanced therapy. Early-phase studies have shown the combination is tolerable and has shown encouraging anti-tumor activity, with partial responses and stable disease observed[1][3][7].
