Overview

Pidacmeran is a self-replicating messenger RNA (mRNA) vaccine developed for active immunization against SARS-CoV-2, the virus responsible for COVID-19. It encodes an extended signal peptide of the SARS-CoV-2 spike glycoprotein, linked at the 3' end by a glycine/serine-rich linker to a trimerization domain from T4 fibritin, and includes untranslated regions and a poly(A) tail. The mRNA incorporates N1-methylpseudouridine instead of uridine to enhance stability and translation efficiency. Pidacmeran was initially developed by BioNTech US and later co-developed with Pfizer and Shanghai Fosun Pharmaceutical. It was administered intramuscularly using lipid nanoparticle delivery technology typical of mRNA vaccines. Its primary indication was prevention of COVID-19 infection; however, its global development status is now discontinued.
