Overview

Pidilizumab (CT-011) is a humanized IgG1 kappa monoclonal antibody that was originally developed as a checkpoint inhibitor targeting Programmed Cell Death Protein 1 (PD-1). However, subsequent biochemical and structural analyses revealed that its primary molecular target is actually Delta-like ligand 1 (DLL1), rather than PD-1. Developed by CureTech and later licensed by Medivation (now part of Pfizer), pidilizumab was designed to enhance the anti-tumor immune response by modulating T-cell and natural killer (NK) cell activity. It has been investigated in Phase II clinical trials for various hematologic malignancies, including diffuse large B-cell lymphoma (DLBCL), follicular lymphoma, and multiple myeloma, as well as for solid tumors such as pediatric high-grade glioma. Despite promising early-phase data, its clinical development has largely stalled following the acquisition of Medivation by Pfizer and the clarification of its mechanism of action.
