Overview

Pidolic acid is a naturally occurring amino acid derivative and a cyclic derivative of glutamic acid. Also known as 5-oxoproline or L-pyroglutamic acid, it is found in various tissues throughout the human body—especially in the brain and skin—and plays roles in several metabolic pathways including the gamma-glutamyl cycle involved in glutathione metabolism[1][2][3][5]. Pidolic acid acts as an intermediate in amino acid metabolism and has been studied for its antioxidant and anti-inflammatory properties. It can form complexes with minerals (as "pidolates") to enhance their bioavailability[1][6]. While not approved as a pharmaceutical drug for any specific indication, it is used as an ingredient in dietary supplements marketed for cognitive enhancement and mineral supplementation (e.g., zinc or magnesium pidolate), as well as cosmetic products due to its humectant properties[2][4][5]. Mechanistically, it may support neurotransmitter regulation and act on the cholinergic system; however, more research is needed to establish clinical benefits[1][2].
