PIEZO-101 is an investigational plasmid DNA-based gene therapy developed by Piezo Therapeutics for the treatment of skin aging and extracellular matrix rejuvenation. It is designed as a biostimulator that restores the expression of key structural proteins within the skin to improve integrity and elasticity. The drug is administered via intradermal injection followed by localized electroporation using the proprietary Piezopen device, which facilitates the cellular uptake of the DNA plasmid. PIEZO-101 is currently being evaluated in a Phase 1 first-in-human clinical trial to assess its safety, tolerability, and pharmacodynamic effects in healthy adult female participants.
