Overview

piggyBac CD19 CAR T cells are an autologous, non-viral chimeric antigen receptor (CAR) T-cell therapy targeting the CD19 antigen. Unlike conventional CAR T-cell products that typically utilize lentiviral or retroviral vectors for gene delivery, this therapy employs the piggyBac transposon system. This system uses electroporation to introduce DNA plasmids (transposon and transposase) into T cells, allowing for the stable integration of the CAR transgene into the host genome. This non-viral approach is hypothesized to reduce manufacturing costs and complexity while potentially offering a safer profile regarding insertional mutagenesis. The therapy is primarily being developed for the treatment of relapsed or refractory CD19-positive B-cell malignancies, including various subtypes of non-Hodgkin lymphoma such as diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma.
