Pigment epithelium-derived factor (PEDF) is a multifunctional 50 kDa secreted glycoprotein belonging to the serine protease inhibitor (serpin) superfamily. Despite its classification as a serpin, it does not exhibit protease inhibitory activity. Instead, PEDF is recognized as one of the most potent endogenous inhibitors of angiogenesis, primarily acting by antagonizing the effects of vascular endothelial growth factor (VEGF) and inducing apoptosis in remodeling endothelial cells. Beyond its anti-angiogenic properties, PEDF provides significant neuroprotection to photoreceptors and motor neurons, exhibits anti-inflammatory effects, and demonstrates anti-tumorigenic activity across various malignancies. Therapeutic development of PEDF includes recombinant protein delivery and gene therapy approaches (using adenoviral or adeno-associated viral vectors) for conditions such as age-related macular degeneration (AMD), diabetic retinopathy, and various cancers.
